Since 1945, the Moscow Nature Reserve consisted of five separate sections located within the Moscow region. The Verkhne-Klyazminsky sector protected the Klyazma glacial moraine plain with meltwater channels; the Verkhne-Moskvoletsky covered the headwaters of the Moskva River. The Gluboko-Istrinsky section included the basin of Lake Glubokoye and moraine hills; the Privolzhsko-Dubnensky preserved ancient dunes in the upper reaches of the Dubna River; and the Prioksko-Terrasny protected the terraces of the Oka River and karst sinkholes.

In 1948, a central European bison nursery was established here. That same year, the sections were reorganized into separate reserves, and in 1951 all of them, except Prioksko-Terrasny, were abolished.
